2026 AACR | Preclinical Data of First-in-Class (FIC) Antibody Drug Conjugate (ADC) IPN60300 Presented

The 2026 American Association for Cancer Research (AACR) Annual Meeting was held in San Diego, USA, from April 17 to 22. Foreseen Biotechnology's strategic partner Ipsen presented two abstracts (Abstract 1665, Abstract 4444) detailing preclinical research data and an abstract (Abstract CT087) on the Phase I/II study design of IPN60300(FS001)1. Foreseen Biotechnology presented an abstract (Abstract 7686) on the discovery of a novel tumor target ITGA2, the antibody target of IPN60300 using the translational-optimized proteomics system (TPS)2.

Preclinical Data of IPN60300

This is a FIC ADC targeting ITGA2, a highly expressed target in various solid tumors such as pancreatic, gastric, and colorectal cancers, while having relatively low expression levels in normal tissues. Preclinical data presented by Ipsen showed that IPN60300 can specifically bind to ITGA2 on the surface of tumor cells and achieve highly efficient endocytosis. Favorable pharmacokinetic profiles, dose -dependent anti-tumor activity and significant tumor tissue-specific enrichment in multiple tumor models were also demonstrated.

Phase I/II Study Design of IPN60300

Ipsen is conducting a Phase I/II first-in-human dose escalation, dose optimization and dose expansion study to evaluate the safety, tolerability, pharmacokinetics, pharmacodynamics, immunogenicity and anti-tumor activity of IPN60300 as a single agent in approximately 114 participants in the US, France and Spain. This Phase I/II clinical trial (NCT07213817)3 is ongoing and an Investigational New Drug (IND) application in China has been accepted by the Center for Drug Evaluation (CDE)4. This marks the first ITGA2-targeted ADC drug to enter into clinical development.

Discovery of Target ITGA2 by Foreseen Biotechnology

Foreseen established a proprietary AI-enabled proteomics platform (Translational-optimized Proteomics System (TPS) with demonstrated capability to accelerate the discovery and validation of FIC targets for drug discovery and diagnostic applications. The abstract titled "Identification of a first-in-class therapeutic target using machine learning enabled translational-optimized proteomics system (TPS)" detailed how ITGA2, the antibody target of IPN60300 (FS001), was discovered and validated. By analyzing a 340-case liver disease progression cohort (encompassing healthy individuals and patients with chronic hepatitis B, liver fibrosis, liver cirrhosis, and hepatocellular carcinoma) and combining it with tissue microarray (TMA) validation, the team screened and identified ITGA2. It is significantly upregulated in hepatocellular carcinoma and overexpressed in pancreatic, gastric, and colorectal cancers, possessing potential to become a broad-spectrum solid tumor target.

From Target Discovery to Global Licensing

IPN60300 (FS001) was created by Foreseen Biotechnology and Escugen Biotechnology. Foreseen discovered the ITGA2 target using the TPS platform and discovered the highly selective and potent antibody targeting ITGA2. Escugen contributed its innovative linker-payload technology. Foreseen advanced IPN60300 (FS001) to IND-enabling stage. In July 2024, Foreseen granted Ipsen an exclusive worldwide license for the development, manufacture and commercialization of IPN60300 (FS001)in a deal with a potential value of up to $1.03 billion USD5, comprising an upfront payment as well as development, regulatory and commercial milestone payments. Foreseen will also be eligible to receive tiered royalties on global sales, subject to successful development and regulatory activities.
